What holds a piece of DNA a thousand times larger than a gene

Sequencing an entire genome — the project that eventually produced the Human Genome Project — meant cloning fragments spanning hundreds of thousands, sometimes millions, of base pairs at a time. Nothing from last lesson comes remotely close to that capacity.

A cosmid vector extends capacity to roughly 45,000 base pairs by combining plasmid elements with a phage packaging signal. Could a similar hybrid strategy be pushed even further, into the hundreds of thousands or millions of base pairs?
